勃林格殷格翰提交Pradaxa辅助药物申请

公司开发的新一代抗凝血药物Pradaxa因其出色的疗效，在上市前就获得了FDA的青睐并被授予突破性药物疗法认证。Pradaxa是一种抗体片段药物，这种药物是通过阻断凝血酶来达到抗凝血的效果。然而，在审批中专家们发现，Pradaxa的出色疗效很有可能超过医生的控制范围而造成血液无法及时凝固，甚至危及患者的生命。直到这时，勃林格殷格翰的工作人员才发现原来有时候疗效太好也不是一件好事。
为了克服Pradaxa的这个副作用,勃林格殷格翰又针对性的开发了一种辅助用药idarucizumab，这种药物能够通过阻断Pradaxa与凝血酶的相互作用而终止前者的抗凝血作用。目前公司已经向美国、加拿大和欧洲的医药管理部门提交了申请。
一直以来，勃林格殷格翰公司在抗凝血药物市场上都处于领先位置，Pradaxa是其新一代的抗凝血药物，目前这种药物的年销售额达到17亿美元，仅次于强生和拜耳的Xarelto和施贵宝和辉瑞的Eliquis。一旦idarucizumab获批上市，Pradaxa必将获得更大的市场份额。同时，也将缓解勃林格殷格翰公司面临的法律纠纷压力，去年公司付出了6亿5千万美元的代价平息了超过4000起与凝血药物副作用有关的法律诉讼。

BoehringerIngelheimsubmittedU.S.,EuropeanandCanadianapplicationsforanantidotetoitsownPradaxa,leadingthewayamongcompaniesdevelopingtreatmentsthatreversetheeffectsofblockbusternext-generationanticoagulants.
Thedrug,whichsecuredtheFDAsbreakthrough-therapydesignation,isanantibodyfragmentdesignedtoreversetheanticoagulanteffectsofPradaxa,whichpreventsclottingbyblockingtheenzymethrombin.Dubbedidarucizumab,Boehringersreversalagentdidjustthatinclinicaltrials,thecompanysaid,chartinganimmediate,completeandsustainedinterruptionofPradaxasmechanismofaction.
Withthefilings,Boehringerleadsitstoprivalinthespace,PortolaPharmaceuticals($PTLA),whichisworkingupanantidotetoawideswathofnewanticoagulants.Thecompanyhasabreakthroughtagofitsownforandexanetalfa,designedtocounteractBayerandJohnson&Johnsons($JNJ)XareltoaswellasBristolMyers-Squibb($BMY)andPfizers($PFE)Eliquis.PortolajustwrappedupitsPhaseIIIprogramandexpectstofileandexanetalfaforFDAapprovalbytheendof2015.
Withitsfirst-moverstatus,BoehringerhopestoboostPradaxasprofileacompetitivefield.ThedrugcurrentlylagsitsXareltoandEliquis,pullinginabout$1.7billionayear,but,iftheFDAapprovesidarucizumab,itllbetheonlynewanticoagulantwithanon-the-marketantidote,likelyaboontosales.

Meanwhile,BoehringerisquicktonotethatthedevelopmentofanantidoteinnowayunderminesitsfaithinPradaxassafety.Lastyear,thecompanyagreedtopayabout$650milliontosettlemorethan4,000lawsuitstiedtothebloodthinner,amovethatcamejustmonthsaftertheFDArevieweddataonthedrugandconcludedthat,whenusedasdirected,itsassafeaswarfarin.
